El trabajo contribuye a explicar el posible uso de los antifolatos para el tratamiento de infecciones por Candida albicans

USO DE ANTIFOLATOS COMO ANTIMICOTICOS: CONEXION ENTRE EL CICLO DEL ACIDO FOLICO Y LA RUTA DE BIOSINTESIS DE ERGOSTEROL EN CANDIDA ALBICANS

A pesar de la amplia utilización de los antifolatos, éstos nunca se encontraron efectivos para el tratamiento de C. albicans. El metotrexato (MTX) muestra alta afinidad por la dihidrofolato reductasa (DHFR) de C. albicans pero no presenta actividad contra el hongo en tratamientos in vivo. En un reciente trabajo comprobamos que la pirimetanina interfiere con la ruta de biosíntesis de ergosterol en C. albicans y que es mucho más activa en presencia de los azoles. Esto ha sacado a la luz el punto de conexión entre el ciclo del ácido fólico y la ruta de biosíntesis del ergosterol. Estos resultados podrían ser de interés para nuevos tratamientos contra C. albicans que incluyan antifolatos o una mezcla de antifolatos y otros antifúngicos.
